Sleep Europe 2026 | Orexin antagonists in RLS: what does the current evidence show?

Diego Garcia-Borreguero, MD, PhD, Sleep Research Institute, Madrid, Spain, discusses the effectiveness of orexin antagonists in treating restless legs syndrome (RLS), highlighting their ability to improve not only sleep but also the core motor sensory disturbances associated with the condition. Dr Garcia-Borreguero notes that the exact mechanism of action is still being explored, but possibilities include orexin’s role in motor control and arousal, with studies suggesting increased orexin levels in patients with RLS in the evening and at night. This interview took place at Sleep Europe 2026, which was held in Maastricht, Netherlands.
